Defensins are small cationic proteins rich in cysteine, found in both vertebrates and invertebrates, and play a crucial role in host defense. They have broad-spectrum antimicrobial activity against bacteria, fungi, and viruses. Beta-defensins, secreted by leukocytes and various epithelial cells, are the most widely distributed. One such peptide, hBD-1 (3.9 kDa, 36 amino acids), consists of a beta sheet with three intramolecular disulfide bonds. It is constitutively produced by epithelial tissues in the urogenital and respiratory tracts and inducible in keratinocytes of human skin by lipopolysaccharides and peptidoglycan. hBD-1 exhibits antimicrobial activity against several pathogenic microorganisms, including E.coli. However, its activity is salt-sensitive and inhibited in a concentration-dependent manner. Furthermore, hBD-1 acts as a chemoattractant for CC chemokine receptor (CCR)6-expressing HEK293 cells, indicating its usage of CCR6 as a receptor.
